ABSTRACT

BACKGROUNDS: With the rapid spread of COVID-19, strict home confinement has been implemented in most parts of Chinese regions. Millions of people were not allowed to leave their homes except for special reasons. Home confinement plays an essential role in curbing pandemic and promoting preventive behaviors, but it may affect individuals' mental health as well. OBJECTS: The objective of this study was to explore the psychological impacts of home confinement. MATERIALS AND METHODS: We collected more than 150,360 Weibo messages from 5,370 Chinese active users, and then extracted psycho-linguistic features from these messages. Psycho-linguistic analysis was carried out using the 2 (confinement vs. non-confinement) × 2 (before vs. after confinement) repeated measure analysis of variance (RM ANOVA). RESULTS: The results showed that the frequency of positive emotion words was remarkably decreased during home confinement [F (1,5368) = 7.926, p = 0.005, η2 = 0.001]. In high-endemic subgroup, home confinement also reduced the frequency of exclusion words [F (1,3445) = 4.518, p = 0.034, η2 = 0.001] and inhibition words [F (1,3445) = 10.154, p = 0.001, η2 = 0.003]. CONCLUSION: Home confinement caused a decline in the use of positive emotion words. This indicates that home confinement can increase the frequency of negative emotions. The changes of exclusion words and inhibition words in high-endemic areas may be related to the high epidemic threat and the urgent need for social distancing in these areas.

ABSTRACT

Lake eutrophication has become a critical environmental issue due to the global effects of anthropogenic activities and climate change, and has been comprehensively studied for many years. A series of models and indicators have been proposed to assess the trophic state of lakes. The trophic state index (TSI) is a synthetic index that integrates chlorophyll-a, water clarity, and total phosphorus and is widely used to evaluate the trophic state of aquatic environments. In this study, we collected in situ lake samples (N = 431) from typical lakes to match Sentinel-2 MultiSpectral Instrument (MSI) imagery data using the Case 2 Regional Coast Color processor. Then we developed a new empirical model, TSI = -34.04 x (band 4/band 5) - 1.114 x (band 1/band 4) + 97.376. This model is valid for all of China, with good performance and few errors (RMSE = 7.36;MAE = 6.25) for the validation dataset. Recognizing that over 94% of the Chinese population located along eastern watersheds and large lakes have competing water uses, and given the TSI model on the seasonal scales, we further estimated the mean TSI and trophic state in eastern Chinese lakes (> 100 km2) from 2019 to 2020. The results revealed that more lakes were eutrophic in autumn (94.28%) than in spring (> 77.14%), indicating a serious eutrophication of eastern lakes. Although the eastern lakes have been studied in more detail, this study found that eutrophication still has markedly negative impacts on lake ecosystems. In addition, no significant improvement was observed in spring, most likely due to the months of curfew/lockdown from January 2020 onwards due to COVID-19. This may be due to the enrichment of nutrients deposited in sediment or watershed soil, which can be characterized as "autochthonous sources" of lake eutrophication, over decades with high rates of economic development. This study demonstrates the applicability of Sentinel-2 MSI data to monitor lake eutrophication as well as the feasibility of blue/red and red/red edge combinations. The framework and TSI model used bands available on MSI sensors to develop a novel approach for generating historical eutrophication data for large-scale evaluation of and decision-making related aquatic environmental changes, even in poorly studied areas.

ABSTRACT

Vaccine uptake is considered as one of the most effective methods of defending against COVID-19 (coronavirus disease 2019). However, many young adults are hesitant regarding COVID-19 vaccines, and they actually play an important role in virus transmission. ABSTRACT

As suicides incurred by the COVID-19 outbreak keep happening in many countries, researchers have raised concerns that the ongoing pandemic may lead to "a wave of suicides" in society. Suicidal ideation (SI) is a critical factor in conducting suicide intervention and also an important indicator for measuring people's mental health. Therefore, it is vital to identify the influencing factors of suicidal ideation and its psychological mechanism during the outbreak. Based on the terror management theory, in the present study we conducted a social media big data analysis to explore the joint effects of mortality salience (MS), negative emotions (NE), and cultural values on suicidal ideation in 337 regions on the Chinese mainland. The findings showed that (1) mortality salience was a positive predictor of suicidal ideation, with negative emotions acting as a mediator; (2) individualism was a positive moderator in the first half-path of the mediation model; (3) collectivism was a negative moderator in the first half-path of the mediation model. Our findings not only expand the application of the terror management theory in suicide intervention but provide some insights into post-pandemic mental healthcare. Timely efforts are needed to provide psychological interventions and counseling on outbreak-caused negative emotions in society. Compared with people living in collectivism-prevailing regions, those living in individualism-prevailing regions may be more vulnerable to mortality salience and negative emotions and need more social attention.

ABSTRACT

With the outbreak of severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2), coronaviruses have begun to attract great attention across the world. Of the known human coronaviruses, however, Middle East respiratory syndrome coronavirus (MERS-CoV) is the most lethal. Coronavirus proteins can be divided into three groups: nonstructural proteins, structural proteins, and accessory proteins. While the number of each of these proteins varies greatly among different coronaviruses, accessory proteins are most closely related to the pathogenicity of the virus. We found for the first time that the ORF3 accessory protein of MERS-CoV, which closely resembles the ORF3a proteins of severe acute respiratory syndrome coronavirus and SARS-CoV-2, has the ability to induce apoptosis in cells in a dose-dependent manner. Through bioinformatics analysis and validation, we revealed that ORF3 is an unstable protein and has a shorter half-life in cells compared to that of severe acute respiratory syndrome coronavirus and SARS-CoV-2 ORF3a proteins. After screening, we identified a host E3 ligase, HUWE1, that specifically induces MERS-CoV ORF3 protein ubiquitination and degradation through the ubiquitin-proteasome system. This results in the diminished ability of ORF3 to induce apoptosis, which might partially explain the lower spread of MERS-CoV compared to other coronaviruses. In summary, this study reveals a pathological function of MERS-CoV ORF3 protein and identifies a potential host antiviral protein, HUWE1, with an ability to antagonize MERS-CoV pathogenesis by inducing ORF3 degradation, thus enriching our knowledge of the pathogenesis of MERS-CoV and suggesting new targets and strategies for clinical development of drugs for MERS-CoV treatment.

ABSTRACT

COVID-19 is mainly associated with respiratory distress syndrome, but a subset of patients often present gastrointestinal (GI) symptoms. Imbalances of gut microbiota have been previously linked to respiratory virus infection. Understanding how the gut-lung axis affects the progression of COVID-19 can provide a novel framework for therapies and management. In this study, we examined the gut microbiota of patients with COVID-19 (n = 47) and compared it to healthy controls (n = 19). Using shotgun metagenomic sequencing, we have identified four microorganisms unique in COVID-19 patients, namely Streptococcus thermophilus, Bacteroides oleiciplenus, Fusobacterium ulcerans, and Prevotella bivia. The abundances of Bacteroides stercoris, B. vulgatus, B. massiliensis, Bifidobacterium longum, Streptococcus thermophilus, Lachnospiraceae bacterium 5163FAA, Prevotella bivia, Erysipelotrichaceae bacterium 6145, and Erysipelotrichaceae bacterium 2244A were enriched in COVID-19 patients, whereas the abundances of Clostridium nexile, Streptococcus salivarius, Coprococcus catus, Eubacterium hallii, Enterobacter aerogenes, and Adlercreutzia equolifaciens were decreased (p < 0.05). The relative abundance of butyrate-producing Roseburia inulinivorans is evidently depleted in COVID-19 patients, while the relative abundances of Paraprevotella sp. and the probiotic Streptococcus thermophilus were increased. We further identified 30 KEGG orthology (KO) modules overrepresented, with 7 increasing and 23 decreasing modules. Notably, 15 optimal microbial markers were identified using the random forest model to have strong diagnostic potential in distinguishing COVID-19. Based on Spearman's correlation, eight species were associated with eight clinical indices. Moreover, the increased abundance of Bacteroidetes and decreased abundance of Firmicutes were also found across clinical types of COVID-19. Our findings suggest that the alterations of gut microbiota in patients with COVID-19 may influence disease severity. Our COVID-19 classifier, which was cross-regionally verified, provides a proof of concept that a set of microbial species markers can distinguish the presence of COVID-19.

During the COVID-19 pandemic, every day, updated case numbers and the lasting time of the pandemic became major concerns of people. We collected the online data (28 January to 7 March 2020 during the COVID-19 outbreak) of 16,453 social media users living in mainland China. Computerized machine learning models were developed to estimate their daily scores of the nine dimensions of the Symptom Checklist-90 (SCL-90). Repeated measures analysis of variance (ANOVA) was used to compare the SCL-90 dimension scores between Wuhan and non-Wuhan residents. Fixed effect models were used to analyze the relation of the estimated SCL-90 scores with the daily reported cumulative case numbers and lasting time of the epidemic among Wuhan and non-Wuhan users. In non-Wuhan users, the estimated scores for all the SCL-90 dimensions significantly increased with the lasting time of the epidemic and the accumulation of cases, except for the interpersonal sensitivity dimension. In Wuhan users, although the estimated scores for all nine SCL-90 dimensions significantly increased with the cumulative case numbers, the magnitude of the changes was generally smaller than that in non-Wuhan users. The mental health of Chinese Weibo users was affected by the daily updated information on case numbers and the lasting time of the COVID-19 outbreak.

ABSTRACT

The global COVID-19 pandemic may significantly affect the experiences of death and bereavement. This study aimed to learn from recent outbreaks of infectious diseases and further understand their impacts on bereavement. We obtained psychological status scores for 32 individuals bereaved due to COVID-19 and 127 individuals bereaved due to non-COVID-19 causes using the online ecological recognition (OER) approach. Next, a sentiment analysis and independent sample t-test were performed to examine the differences between these two groups. The results indicated that the individuals bereaved due to COVID-19 were more insecure and more preoccupied with the grief of the moment than those bereaved due to non-COVID-19 reasons, while the latter group had higher depression scores than the former group. This study can guide policy-makers and clinical practitioners to provide more targeted and sustainable post-bereavement support for both bereaved groups during the COVID-19 period.

Background: Coronavirus disease 2019 (COVID-19) threatens people's physical and mental health, globally, and it may even trigger suicide ideation and suicidal behavior. Aims: We aimed to examine the impact of COVID-19 on suicide risk by sampling Chinese Weibo users and analyzing their social media messages. Method: We predicted the probability of suicide (including hopelessness, suicidal ideation, negative self-evaluation, and hostility) of Weibo users in order to assess the changes in suicide probability at different times. Repeated-measures ANOVA was performed to examine the differences in suicide probability in different regions during different periods. Results: There was no significant difference in suicide probability between profoundly infected areas (PIAs) and less infected areas (LIAs) before the outbreak of COVID-19. LIAs had an increase in hopelessness during the COVID-19 growth period, while hopelessness and hostility in PIA increased during the COVID-19 decline period, indicating potential suicide probability. Limitations: Results should be interpreted with caution, and cross-cultural research may be considered in the future. Conclusion: COVID-19 has a dynamic impact on suicide probability. Using data from online social networks may help to understand the impact pattern of COVID-19 on people's suicide probability.

ABSTRACT

SARS-CoV-2 is the etiological agent responsible for the ongoing pandemic of coronavirus disease 2019 (COVID-19). The main protease of SARS-CoV-2, 3CLpro, is an attractive target for antiviral inhibitors due to its indispensable role in viral replication and gene expression of viral proteins. The search of compounds that can effectively inhibit the crucial activity of 3CLpro, which results to interference of the virus life cycle, is now widely pursued. Here, we report that epigallocatechin-3-gallate (EGCG), an active ingredient of Chinese herbal medicine (CHM), is a potent inhibitor of 3CLpro with half-maximum inhibitory concentration (IC50) of 0.874 ± 0.005 µM. In the study, we retrospectively analyzed the clinical data of 123 cases of COVID-19 patients, and found three effective Traditional Chinese Medicines (TCM) prescriptions. Multiple strategies were performed to screen potent inhibitors of SARS-CoV-2 3CLpro from the active ingredients of TCMs, including network pharmacology, molecular docking, surface plasmon resonance (SPR) binding assay and fluorescence resonance energy transfer (FRET)-based inhibition assay. The SPR assay showed good interaction between EGCG and 3CLpro with KD ~6.17 µM, suggesting a relatively high affinity of EGCG with SARS-CoV-2 3CLpro. Our results provide critical insights into the mechanism of action of EGCG as a potential therapeutic agent against COVID-19.

BACKGROUND: During the COVID-19 pandemic, residential lockdowns were implemented in numerous cities in China to contain the rapid spread of the disease. Although these stringent regulations effectively slowed the spread of COVID-19, they may have posed challenges to the well-being of residents. OBJECTIVE: This study aims to explore the effects of residential lockdown on the subjective well-being (SWB) of individuals in China during the COVID-19 pandemic. METHODS: The sample consisted of 1790 Sina Weibo users who were residents of cities that imposed residential lockdowns, of which 1310 users (73.18%) were female, and 3580 users who were residents of cities that were not locked down (gender-matched with the 1790 lockdown residents). In both the lockdown and nonlockdown groups, we calculated SWB indicators during the 2 weeks before and after the enforcement date of the residential lockdown using individuals' original posts on Sina Weibo. SWB was calculated via online ecological recognition, which is based on established machine learning predictive models. RESULTS: The interactions of time (before the residential lockdown or after the residential lockdown) × area (lockdown or nonlockdown) in the integral analysis (N=5370) showed that after the residential lockdown, compared with the nonlockdown group, the lockdown group scored lower in some negative SWB indicators, including somatization (F1,5368=13.593, P<.001) and paranoid ideation (F1,5368=14.333, P<.001). The interactions of time (before the residential lockdown or after the residential lockdown) × area (developed or underdeveloped) in the comparison of residential lockdown areas with different levels of economic development (N=1790) indicated that the SWB of residents in underdeveloped areas showed no significant change after the residential lockdown (P>.05), while that of residents in developed areas changed. CONCLUSIONS: These findings increase our understanding of the psychological impact and cost of residential lockdown during an epidemic. The more negative changes in the SWB of residents in developed areas imply a greater need for psychological intervention under residential lockdown in such areas.

Abstract COVID-19 (Corona Virus Disease 2019) has attacked many countries around world and caused profound impacts on public life. The outbreak of pandemic and other relevant factors are considered to cause emotion responses of residents. And the emotion responses of individuals are crucial for the execution of the prevention and control measures. By analyzing the linguistic features of posts on social media, this study aims to explore the change of public emotion responses during COVID-19 in China. We sampled 22,423 Weibo users and collected their Weibo posts by provincial area each day from January 1st, 2020 to April 18th, 2020. Next, we extracted linguistic features from posts according to the emotion-related dictionary. Based on important news and information released by the national and international organizations of public health, we divided the period from January 1st, 2020 to April 18th, 2020 into four stages (the initial period, the outbreak period, the stable period, and the prevention and control period). Then we gathered linguistic features by stage. After that, ANOVA was performed to examine the differences among these four stages. The results showed that the frequencies of 11 word categories showed significant differences among four stages, including fear, disappointment, guilt, missing, anger, panic, blessing, faith, love, praise, and surprise. The uses of several negative emotion words, such as fear, disappointment, guilt, and anger, increased saliently in the outbreak period compared with the initial period. Besides, panic words decreased significantly in the prevention and control period compared with the outbreak period. However, missing words were used more in the prevention and control period than other three periods. Moreover, people expressed more faith words and less love words in the outbreak period than the initial periods. Besides, people used more blessing words in the outbreak period compared with the stable period and prevention and control period. And praise words were used more in the outbreak period and the stable period compared with the initial period. The frequency of surprise words was significantly low only in the initial period. This study contributed to the understanding of public emotion responses during COVID-19, and had implications for the evidence-based execution of prevention and control measures.

ABSTRACT

Coronavirus disease 2019 (COVID-19) with the infection of SARS-CoV-2 has become a serious pandemic worldwide. However, only few studies focused on risk factors of prolonged SARS-CoV-2 RNA detection among patients with COVID-19. We included 206 adult patients with laboratory-confirmed COVID-19 from two hospitals between 23 Jan and 1 April 2020. Least absolute shrinkage and selection operator (LASSO) analysis was used to screen out independent risk factors of SARS-CoV-2 RNA detection. By multivariate binomial logistic regression analysis and Cox regression analysis, we further determined the associations between SARS-CoV-2 RNA detection and potential risk factors. All patients had two negative SARS-CoV-2 tests with 33 days of median duration of SARS-CoV-2 RNA detection (interquartile range: 25.2-39 days). LASSO and binomial logistic regression analyses suggested that delayed hospital admission (adjusted OR = 3.70, 95% CI: 1.82-7.50), hypokalemia, and subpleural lesion (adjusted OR = 4.32, 95% CI: 1.10-16.97) were associated with prolonged SARS-CoV-2 RNA detection. By LASSO and multivariate Cox regression analyses, we observed that delayed hospital admission, subpleural lesion, and high-dose corticosteroid use were independent risk factors of prolonged SARS-CoV-2 RNA detection. Early hospital admission shortened 5.73 days of mean duration of SARS-CoV-2 RNA detection than delayed hospital admission after adjusting confounding factors. Our study demonstrated that delayed hospital admission and subpleural lesion were associated with prolonged SARS-CoV-2 RNA detection among patients with COVID-19. The use of high-dose corticosteroids should be interpreted with extreme caution in treating COVID-19.

ABSTRACT

Coronavirus disease 2019 (COVID-19) is a highly infective disease caused by the severe acute respiratory syndrome coronavirus 2 virus (SARS-CoV-2). Previous studies of the COVID-19 pneumonia outbreak were based on information from the general population. Limited data are available for hemodialysis patients with COVID-19 pneumonia. This report describes the clinical characteristics of COVID-19 in an in-center hemodialysis patient, as well as our experience in implementing steps to prevent the spread of COVID-19 pneumonia among in-center hemodialysis patients. The diagnosis, infection control, and treatment of COVID-19 in hemodialysis patients are discussed in this report, and we conclude with recommendations for how a dialysis facility can respond to COVID-19 based on our experiences.

ABSTRACT

The study aims to understand Twitter users' discourse and psychological reactions to COVID-19. We use machine learning techniques to analyze about 1.9 million Tweets (written in English) related to coronavirus collected from January 23 to March 7, 2020. A total of salient 11 topics are identified and then categorized into ten themes, including "updates about confirmed cases," "COVID-19 related death," "cases outside China (worldwide)," "COVID-19 outbreak in South Korea," "early signs of the outbreak in New York," "Diamond Princess cruise," "economic impact," "Preventive measures," "authorities," and "supply chain." Results do not reveal treatments and symptoms related messages as prevalent topics on Twitter. Sentiment analysis shows that fear for the unknown nature of the coronavirus is dominant in all topics. Implications and limitations of the study are also discussed.

ABSTRACT

The novel coronavirus severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has spread globally to over 200 countries with more than 23 million confirmed cases and at least 800,000 fatalities as of 23 August 2020. Declared a pandemic on March 11 by World Health Organization, the disease caused by SARS-CoV-2 infection, called coronavirus disease 2019 (COVID-19), has become a global public health crisis that challenged all national healthcare systems. This review summarized the current knowledge about virologic and pathogenic characteristics of SARS-CoV-2 with emphasis on potential immunomodulatory mechanism and drug development. With multiple emerging technologies and cross-disciplinary approaches proving to be crucial in our global response against COVID-19, the application of PROteolysis TArgeting Chimeras strategy, CRISPR-Cas9 gene editing technology, and Single-Nucleotide-Specific Programmable Riboregulators technology in developing antiviral drugs and detecting infectious diseases are proposed here. We also discussed the available but still limited epidemiology of COVID-19 as well as the ongoing efforts on vaccine development. In brief, we conducted an in-depth analysis of the pathogenesis of SARS-CoV-2 and reviewed the therapeutic options for COVID-19. We also proposed key research directions in the future that may help uncover more underlying molecular mechanisms governing the pathology of COVID-19.
